I have a 2001 xc70 with 160k I plan on doing a drain and fill on. I am curious as to upon the refill, if I need to cycle the shifter from P to N- to R to D, and then put it in manual and shift up and down from 1st to third?
Also, when draining and replacing, should the fluid be hot coming out...and if so how much to underfill to compensate for thermal expansion?
If anyone has done this, please let me know, these are the only items I am fuzzy on...
Thanks!

No need to do any shifter motions.
The dipstick has markings on both sides. go to lower end of the hot (higher) scale.
Full on the hot scale would be for hot fluid from a lengthy drive.

Yes, it is generally best to change any oil when warm so it drains faster and more completely.
All auto transmissions I know of are checked when running on level ground. The torque converter can't be assured to be full without the engine running.
